I googled it:
Under each user's name you will see buttons called "Exalt" and "Smite". Exalt is used when you'd like to praise another user for a particularly helpful or funny posting. Smite is used when you'd like to smack them, instead! Every "exalt" increases the user's karma rating by one point. Conversely, every smite decreases the karma rating. After exalting or smiting another user, you will have to wait about an hour for your buttons to reappear. The delay is to keep people from going "exalt-crazy". It's tempting to over-exalt your friends, but remember if it's not used judiciously, the karma rating will sort of lose its impact!

I wonder if you can have a negative karma? Yes if you smite them enough. You may only exalt or smite a person once per hour but if you're persistent that means you can give a person -24 karma every day. Pretty mean though.
